Historicamente a piscicultura no estado de Santa Catarina baseia se no cultivo de espécies exóticas como as carpas: comum e chinesas, trutas e a tilápia do Nilo. Isto ocorreu principalmente pela falta de tecnologias para cultivo de espécies nativas. O conhecimento mais aprofundado de técnicas de controle da sexualidade de peixes nativos é um passo importante para inserir essas novas tecnologias e incentivar o cultivo. O jundiá, Rhamdia quelen, é uma espécie nativa, presente na região Sul do Brasil e que vem sendo considerado um peixe promissor para a piscicultura comercial. Isto se deve não só pelo seu filé sem espinhos e de ótima qualidade, mas também pelo seu rápido crescimento chegando a atingir 1000 gramas as fêmeas e 700 gramas os machos em um ciclo de 8 meses de cultivo. O experimento teve como objetivo principal analisar comparativamente diferentes metodologias da produção de monosexo, gerando um protocolo para feminilização de jundiá Rhamdia quelen. As metodologias aplicadas para reversão sexual do jundiá foram o arraçoamento com hormônio feminilizante e o banho de imersão com o mesmo hormônio. Para o primeiro tratamento foram selecionadas 1000 larvas para as 2 diferentes concentrações de 17β-estradiol incorporada à ração (105 mg/kg e 115 mg/kg). A dieta com hormônio se estendeu por 30 dias, e após este período, as larvas passaram a ser alimentadas com ração sem hormônio. Para o segundo tratamento também foram selecionadas 1000 larvas para as diferentes fases ontogênicas: 5, 10 e 18DPE (dias pós-eclosão) do jundiá. Os banhos de imersão foram feitos com 3 mg/L de 17β-estradiol por 36h, e passado este período o nível de água nos tanques e a renovação foram retornado ao normal. Os resultados obtidos para o primeiro tratamento de feminilização foram de 100% e 87,5% para as concentrações de 105mg/kg e 115mg/kg de 17β-estradiol, respectivamente. Para o tratamento de banho de imersão os resultados obtidos foram de 97,5% para 5 DPE; 95,12% para 10 DPE; e 92,5% para 18 DPE. A partir dos resultados obtidos, sugere-se 105 mg/kg de hormônio incorporado a ração e a fase ontogênica de 5 DPE como os melhores tratamentos para o protocolo de feminilização. Epagri is carrying out a project aimed at the induction of mutants for resistance to banana diseases by
irradiation of in vitro shoots of the cultivar SCS451 Catarina. The first step was the determination of
sensitivity to gamma rays using doses of 0; 12.5; 25; 37.5 and 50Gy. As it is known if in a shoot tip
after treatment with any mutagen the mutation may be induced in M1V1, however, there is the
occurrence of chimerism and further subcultures (M1V2, M1V3, M1V4, etc.) are needed to select and
isolate the stable mutants (periclinal chimeras or solid mutants). The shoots used were cultured using
the appropriate multiplication and growth media for banana. The experimental unit was composed of 3
flasks in M1V1 and 5 flasks in the others (M1V2 to M1V4) and 5 shoots were always inoculated in each
flask. The mean height and shoot regeneration rate (number of shoots formed at the number of
inoculated shoots) were evaluated at the end of each subculture (30-40 days interval). The treatments
that received the two highest doses (37,5 and 50Gy) presented the lowest regeneration rate (1,91 and
1,48, respectively) and the shortest shoot height (2,09 and 1,72 cm, respectively) at the end of M1V1.
However, with the advance of the subcultures, the results of these treatments were like the others, so
that in M1V4 there were no significant differences. As in mutation breeding whenever possible it is
recommended to use more than one dose, based on the obtained results it was concluded that these two
doses (37,5 and 50Gy) can be used for the continuation of the research. For the continuation of the work
shoots were irradiated with each of these doses and the material is being led up to the M1V4 generation
